Sounds like a permissions issue. Try running as admin?

It appears that the tweaker is trying to modify the HOSTS file ('C:\Windows\system32\drivers\etc\HOSTS), which occurs when you're using cyberkitsune's proxy. The question is: are you using cyberkitsune's proxy?

If so, then maybe you're not running tweaker as an admin. Try that.
If not, then deactivate the proxy plugin in the tweaker. Go to the Orb menu, select the "Plugins" button, deactivate the PSO2proxy plugin and save.

Be aware, there are other problems right now: http://www.pso-world.com/forums/showthread.php?t=231790, so you should wait until the problem is settled.
