Online is completely separate from offline aside from levels. But it won't unlock offline missions/difficulties.
I'd imagine you could level yourself up like crazy before starting story like that. Though if I'm wrong feel free to correct me.
Online is just missions to level-up and get items pretty much. You cannot unlock anything for offline playing only online; however, you can essentially make offline mode prior to beating the story a complete joke by overleveling online and mking yourself able to waste everything you face without the slightest of difficulty. You also cannot HOST any missions or field runs in any area you have not unlocked in offline mode as well, so if you host with a fresh new character, you will only have the first three areas unlocked to play, and must beat the rest of the areas' story missions before they become unlocked for online mode (however, the loophole around this is to have someone else who HAS unlocked every area to host for you, since it's based on the host's cartridge).
