Which classes do people use for challenge mode generally? Is it best to have a diverse class line-up, or is one clearly better based on the items they're given?
Back from when I did it, Hunter and Ranger were the most useful. I think it was more of the Photon Arts that were available for the Hunter and keeping chains alive for the Ranger. I avoid using the other classes, Hunter and Ranger have more oomph than the other two classes at the moment. Having Diga is nice for a stage or two where you need to push a cart though.
With that said, as long as you the skill you can get the job done with any class either way.

To further develop, even though Joshboyd1209 answered most of the question:
The % increases the weapon's TEC if, and ONLY if you use the matching technique on an enemy weak to it. So, let's say your weapon has 500 TEC, is a rod with 4 techniques on it, has 60% , you have 1000 natural TEC and you use Damgrants level 30 (100% TEC mod) on a enemy with 500 MND. The damage formula would be:
(([1000+(500x (1+0.30+0.60+0.20))] X 100%) - 500 ) / 5 = 310 damage
As you can see, on top of the default 30% added to the weapon's TEC for using the proper element, your weapon's elemental % boosts your weapon's base TEC as well.
As for empower, it is factored at the end of the damage formula, affects the damage as long as a technique of the matching element is casted and varies depending on the weapon type. IIRC, wands have empower bonuses ranging from +10% to +20%, Rods range from 10% to 30%, while Madoogs...I won't bother since they'll never receive any love from me (Except the Dol Vaverg, because it has a fashionable scarf).
Anyways, using the same scenario as above, except that your weapon is , and has an empower bonus of 30%:
[(([1000+(500 x (1+ 0.3))] X 100%) - 500 ) / 5 ] x 120% = 276 damage
Obviously, even though the empower bonus always works, you'd prefer to have it on top of a weapon with the matching element for maximum damage. If we used the first example, for instance, your damage would have been around 400-ish.
