Red Boxes are rare items in the game. When you see a red box, it means the item is rare. It can be dropped from almost any enemy.

The contents of a Red Box varies as well...grab these no matter what. Drop whatever you have to in your inventory to make room for it..

Hahahaha. Sorry, I misinterpreted. I thought you meant like a blue box that you break to get an item. Heh, my bad. Thx.
